目的探讨转录沉默子8(TIS8)基因在调控肝再生进程中的作用。方法野生小鼠(WT鼠)随机分为70%肝切除术组(PH组,每小组5只)和假手术组(SH组,分组同前)。TIS8基因敲除鼠(TIS8鼠)亦实施PH,随即分为术后48h组和术后72h组,每组5只。使用实时RT-PCR法测定PH和SH后WT鼠不同时相肝组织TIS8的mRNA水平;Western blot检测PH后48 h和72h WT鼠和TIS8鼠肝组织中TIS8蛋白质的表达水平并结合肝细胞复制周期进行比较。结果WT鼠TIS8基因的诱导表达始于PH后6~12h,继而下调,但于48h表达再次上调。WT鼠TIS8蛋白质的表达存在于肝静止期,PH后2~6h表达减弱,但12-36h表达再次升高,而48~72h表达略减弱;然而,TIS8鼠术后48h和72 h TIS8蛋白质的表达均明显减弱(P<0.01),说明,肝再生过程中TIS8蛋白质表达的时段恰好对应于肝细胞的有丝分裂进程。结论正常肝再生的有效进程中,TIS8的表达是必需的;TIS8参与了肝细胞有丝分裂进程的调控。
Objective To investigate the role of transcription silencer 8 (TIS8) gene in the regulation of liver regeneration. Methods Wild mice (WT mice) were randomly divided into 70% hepatectomy group (PH group, 5 mice per group) and sham operation group (SH group, the same as above). TIS8 knockout mice (TIS8 mice) also performed PH, then divided into postoperative 48h group and postoperative 72h group, 5 in each group. Real-time RT-PCR was used to determine the TIS8 mRNA level in the liver tissue of WT mice at different time points. The expression of TIS8 protein in liver tissue of WT and TIS8 mice at 48 h and 72 h after PH treatment was detected by Western blot. Cycle to compare. Results The induced expression of TIS8 gene in WT mice began at 6-12 h after PH, then down-regulated, but up-regulated at 48 h. The expression of TIS8 protein in WT mice was in quiescent phase of liver. The expression of TIS8 protein decreased from 2 to 6 hours after PH, but increased again from 12 to 36 hours and slightly decreased from 48 to 72 hours. However, TIS8 protein (P <0.01), indicating that the period of TIS8 protein expression during liver regeneration corresponds exactly to the mitotic progression of hepatocytes. Conclusion The expression of TIS8 is necessary in the process of normal liver regeneration. TIS8 is involved in the regulation of mitosis in hepatocytes.
