目的观察在大鼠实验性脑出血(ICH)后不同时间点血肿周围白细胞介素-1β(IL-1β)表达情况及凝血酶对其影响。方法采用自体血、凝血酶注入法分别制作大鼠模型,应用免疫组化法观察不同时间点各组大鼠IL-1β的表达情况及凝血酶对其表达的影响。结果在造模后1h,脑出血组和凝血酶组病灶周围即可见IL-1β表达,6h达高峰,以后逐渐下降,48h阳性细胞基本消失。在相应时间点,ICH+水蛭素组血肿周围IL-1β的表达水平均明显低于ICH组(P<0.01),且细胞着色浅,凝血酶+水蛭素组病灶周围IL-1β的表达水平均明显低于凝血酶组(P<0.01),且细胞着色浅。假手术组仅有少量阳性细胞表达。结论IL-1β参与了ICH后脑损伤,凝血酶可能通过促进IL-1β的表达而发挥脑损伤作用。
Objective To observe the expression of interleukin-1β (IL-1β) around the hematoma and the effect of thrombin on the rats at different time points after experimental intracerebral hemorrhage (ICH). Methods The rat model was established by autologous blood and thrombin injection. The expression of IL-1β and the effect of thrombin on the expression of IL-1β at different time points were observed by immunohistochemistry. Results The expression of IL-1β was observed around the lesion in cerebral hemorrhage group and thrombin group at 1 hour after model establishment, reaching the peak at 6 hours and then gradually decreasing. The positive cells disappeared at 48 hours. At corresponding time points, the expression level of IL-1β in ICH + hirudin group was significantly lower than that in ICH group (P <0.01), and the cells were stained lightly and the expression level of IL-1β in thrombin + hirudin group was significantly Lower than the thrombin group (P <0.01), and the cells were colored light. Only a few positive cells were expressed in sham operation group. Conclusion IL-1βis involved in brain injury after ICH. Thrombin may play a role in brain injury by promoting the expression of IL-1β.
