用硫酸铵分级后再用phenyl-Sepharose CL-4B,DEAE-Sephadex A-50柱层析得到均一的人红细胞谷胱甘肽过氧化物酶(GSH-Px)。该酶在SDS聚丙烯酰胺凝胶电泳呈单一区带。用酶联免疫吸附法测定了克山病病区儿童和非病区儿童血GSH-Px活力;同时测定了大骨节病区和非病区儿童血GSH-Px活力。实验结果表明病区明显低于非病区儿童。
After being fractionated with ammonium sulfate, homogeneous human erythrocyte glutathione peroxidase (GSH-Px) was obtained by column chromatography on a phenyl-Sepharose CL-4B, DEAE-Sephadex A-50 column. The enzyme showed a single band on SDS polyacrylamide gel electrophoresis. The serum GSH-Px activities in children and non-ward children with Keshan disease were detected by enzyme-linked immunosorbent assay (ELISA). The GSH-Px activities in blood of KBD patients and non-disease children were also measured. The experimental results show that the ward is significantly lower than non-ward children.
