目的:观察丹酚酸胶囊对Beagle犬急性心肌缺血模型缺血心肌的保护作用。方法:连续2周每日给予Beagle犬25mg.kg-1、50mg.kg-1、100mg.kg-1丹酚酸胶囊后,观察二步法结扎冠状动脉致Beagle犬急性心肌缺血模型的心率(HR)、血压(MAP)、心肌血流量(MBF)和冠脉阻力(CVR)的变化,以及血清肌酸激酶同工酶(CK-MB)、丙二醛(MDA)含量、过氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)活性的变化。结果给予高、中、低剂量丹酚酸胶囊组可明显降低HR、CVR、CK-MB和MDA含量(P<0.05,P<0.01),显著提高MBF、SOD活性和GSH-PX活性(P<0.05,P<0.01),对血压无明显影响(P>0.05)。结论:丹酚酸胶囊可有效降低Beagle犬急性心肌缺血程度,改善心肌供血,提高其自由基清除能力和抗氧化能力,对犬的缺血心肌具有保护作用。
Objective: To observe the protective effects of salvianolic acid capsules on ischemic myocardium in beagle dogs with acute myocardial ischemia. Methods: Beagle dogs were treated with 25mg.kg-1, 50mg.kg-1 and 100mg.kg-1 salvianolic acid capsules daily for 2 weeks. The heart rate of Beagle dogs with acute myocardial ischemia induced by two-step coronary artery ligation (HR), blood pressure (MAP), myocardial blood flow (MBF) and coronary artery resistance (CVR), as well as serum creatine kinase MB, MDA, Changes in the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX). Results High, medium and low doses of salvianolic acid capsules could significantly reduce the contents of HR, CVR, CK-MB and MDA (P <0.05, P <0.01) and significantly increase the activity of MBF and SOD and the activity of GSH-PX 0.05, P <0.01), no significant effect on blood pressure (P> 0.05). Conclusion: Salvianolic acid capsules can effectively reduce acute myocardial ischemia in Beagle dogs, improve myocardial blood supply, improve their free radical scavenging ability and antioxidant capacity, and have a protective effect on canine ischemic myocardium.
