目的研究抑癌基因线粒体琥珀酸脱氢酶(SDH)及其表达产物SDH蛋白在正常肾上腺髓质和肾上腺嗜铬细胞瘤中的表达,比较SDH mRNA的表达与SDH蛋白的表达关系。方法取正常肾上腺髓质和嗜铬细胞瘤组织标本,应用逆转录——聚合酶链反应(RT-PCR)及免疫组织化学方法检测SDH mRNA及SDH蛋白的表达情况。结果RT-PCR显示,嗜铬细胞瘤组织和正常肾上腺髓质均可扩增出灰白色的SDH条带。免疫组化显示SDH蛋白在嗜铬细胞瘤中表达为75.8,在正常肾上腺髓质表达为10。结论肾上腺嗜铬细胞瘤存在SDH基因突变,推测SDH基因与嗜铬细胞瘤的发生存在明显相关性。
Objective To study the expression of mitochondrial superoxide dismutase (SDH) and its expression product SDH in normal adrenal medullary pheochromocytoma and adrenal pheochromocytoma, and to compare the expression of SDH mRNA and SDH protein. Methods Normal adrenal medulla and pheochromocytoma tissues were obtained. The expressions of SDH mRNA and SDH protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry. Results RT-PCR showed that gray-scale SDH bands were amplified in both pheochromocytoma and normal adrenal medulla. Immunohistochemistry showed SDH protein was expressed as 75.8 in pheochromocytoma and 10 in normal adrenal medulla. Conclusion SDH gene mutation exists in adrenal pheochromocytoma. It is speculated that there is a significant correlation between SDH gene and the occurrence of pheochromocytoma.
