研究通过静态吸附-解吸试验从5种树脂中选择最适合绿原酸纯化的大孔树脂,并通过单因素试验优化动态吸附-解吸条件。结果表明,AB-8型树脂对绿原酸有较高的吸附率和解吸率,优化后的吸附条件为上样流速2mL/min;上样液浓度2 mg/mL;pH 3;上样体积5 BV;解吸条件为洗脱液流速2 mL/min;洗脱液体积2 BV;洗脱液浓度为30%的乙醇,纯化后绿原酸的纯度为76.5%。HPLC-MS分析表明,绿原酸是咖啡酸和奎尼酸缩合的缩酚酸。
The best macroporous resin purified from five kinds of resins was selected by static adsorption-desorption test, and the dynamic adsorption-desorption conditions were optimized by single factor test. The results showed that AB-8 resin had higher adsorption rate and desorption rate for chlorogenic acid. The optimal adsorption conditions were as follows: flow rate of sample 2 mL / min; concentration of sample 2 mg / mL; pH 3; volume of sample 5 BV. The desorption conditions were eluant flow rate 2 mL / min; eluent volume 2 BV; eluent concentration 30% ethanol; purity of chlorogenic acid after purification was 76.5%. HPLC-MS analysis showed that chlorogenic acid is a condensed phenolic acid between caffeic acid and quinic acid.
