目的:建立同时测定大鼠血浆中盐酸小檗碱、大黄素、大黄酚和大黄素甲醚含量的方法。方法:采用高效液相色谱串联质谱法,色谱柱为Shim Pack VP-ODS C_(18)色谱柱(150 mm×2.0 mm,5μm),柱温30℃,流动相为乙腈:0.5%甲酸水溶液(80:20,氨水调pH=3.5),流速0.2ml·min~(-1)。结果:盐酸小檗碱、大黄素、大黄酚、大黄素甲醚的最低检测限范围为1.0~1.2ng·ml~(-1),定量下限范围为5.0~11.5 ng·ml~(-1),日内、日间RSD<2.98%。结论:该法准确、快速,可用于上述4种成分的药动学研究。
Objective: To establish a method for the simultaneous determination of berberine hydrochloride, emodin, chrysophanol and physcion in rat plasma. METHODS: High performance liquid chromatography tandem mass spectrometry (HPLC-MS / MS) was performed on a Shim Pack VP-ODS C 18 column (150 mm × 2.0 mm, 5 μm) with a mobile phase of acetonitrile 0.5% 80:20 ammonia water to adjust pH = 3.5), the flow rate of 0.2ml · min ~ (-1). Results: The detection limit of berberine hydrochloride, emodin, chrysophanol and physcion was 1.0 ~ 1.2ng · ml -1 and the lower limit of quantification was 5.0 ~ 11.5 ng · ml -1 , Day, day RSD <2.98%. Conclusion: This method is accurate and rapid and can be used for pharmacokinetic studies of the above four components.
