目的探讨Parkin基因mRNA及其蛋白在肝细胞癌中的表达规律及其临床意义。方法应用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学(S-P)法检测58例肝细胞癌中Parkin基因mRNA及蛋白的表达情况,结合病人的临床病理资料分析Parkin基因在肝细胞癌中的意义。结果肝癌组织中Parkin基因mRNA及蛋白的表达明显降低,阳性率分别为22.4%(13/58)和25.9%(15/58),明显低于癌旁肝组织的表达阳性率:94.8%(55/58)和89.7%(52/58),其差异有统计学意义(P<0.05),且Parkin基因蛋白表达的缺失与病人的性别、肿瘤的大小、甲胎蛋白(AFP)、HBsAg、以及有无肝硬化无关(P>0.05),而与肿瘤的分化程度、有无包膜及有无门静脉癌栓有关(P<0.05)。结论Parkin基因在肝细胞癌的发生过程中可能起重要的作用,其表达水平可能成为肝细胞癌诊断及判断其生物学特性的重要分子生物学指标。
Objective To investigate the expression of Parkin mRNA and its protein in hepatocellular carcinoma and its clinical significance. Methods The expressions of Parkin mRNA and protein in 58 cases of hepatocellular carcinoma were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry (SP). The positive rates of Parkin mRNA and protein were 22.4% (13/58) and 25.9% (15/58), respectively, which were significantly lower than those in adjacent noncancerous liver tissues. 94.8% (55/58) and 89.7% (52/58), respectively. There was a significant difference between the two groups (P <0.05). The loss of Parkin gene expression was related to the gender, tumor size, AFP, HBsAg, and liver cirrhosis (P> 0.05), but not with the degree of tumor differentiation, with or without capsule and portal vein tumor thrombus (P <0.05). Conclusions Parkin gene may play an important role in the development of hepatocellular carcinoma. The expression level of Parkin gene may be an important molecular biological indicator for the diagnosis of hepatocellular carcinoma and its biological characteristics.
