目的:建立同时测定泸州龙眼肉、核、皮中没食子酸和鞣花酸含量的HPLC方法。方法:依利特Sino Chrom ODS-BP(4.6 mm×250 mm,5μm),流动相乙腈-0.1%磷酸溶液,梯度洗脱,改变波长测定(0~11.5 min,215 nm,测定没食子酸;11.5~25 min,255 nm,测定鞣花酸),柱温30℃,流速1 mL.min-1,进样量10μL。结果:没食子酸在0.229~2.29μg,鞣花酸在0.412 5~4.125μg峰面积与进样量有良好的线性关系(r=0.999 9),平均回收率(n=9)分别为98.27%(RSD 0.63%),98.31%(RSD 0.88%)。结论:该方法灵敏、准确、重复性好,可用于泸州龙眼中没食子酸和鞣花酸的含量测定。
Objective: To establish a HPLC method for the simultaneous determination of gallic acid and ellagic acid in the meat, kernel and skin of Luzhou longan. Methods: The elution was carried out with gradient elution with acetonitrile-0.1% phosphoric acid solution (4.6 mm × 250 mm, 5 μm) 25 min, 255 nm, determination ellagic acid), column temperature 30 ℃, flow rate 1 mL.min-1, injection volume 10μL. Results: The linear range was 0.229 ~ 2.29μg for gallic acid and 0.412 5 ~ 4.125μg for ellagic acid. The average recoveries (n = 9) were 98.27% (r = 0.999 9) RSD 0.63%), 98.31% (RSD 0.88%). Conclusion: The method is sensitive, accurate and reproducible. It can be used for the determination of gallic acid and ellagic acid in Luzhou longan.
