探讨羊栖菜多糖、姬松茸多糖对高糖培养的胰岛β细胞的保护作用及其机制。将胰岛β细胞随机分为5组:正常对照组、模型组、SFPS组、ABP组、两药联合组。应用MTT法检测对胰岛β细胞增殖能力的影响,应用荧光显微镜检测胰岛β细胞凋亡情况,并用RT-PCR法检测bax、caspase-3基因表达量的变化。结果表明,SFPS、ABP能明显减轻高糖对胰岛βTc3细胞的生长抑制,抑制高糖诱导的胰岛β细胞凋亡,并明显降低bax、caspase-3基因表达(p<0.01),两药联合作用更明显,其作用机制可能与调控bax、caspase-3基因表达有关。
To explore the protective effect of polysaccharides of Sargassum fusiforme and Agaricus blazei on islet β cells cultured in high glucose and its mechanism. The islet β cells were randomly divided into 5 groups: normal control group, model group, SFPS group, ABP group, two drug combination group. The effects of MTT assay on the proliferation of pancreatic β cells were observed. The apoptosis of pancreatic β cells was detected by fluorescence microscopy. The expression of bax and caspase-3 genes was detected by RT-PCR. The results showed that SFPS and ABP could significantly reduce the growth inhibition of pancreatic βTc3 cells induced by high glucose and inhibit the apoptosis of pancreatic β-cells induced by high glucose, and significantly reduce the expression of bax and caspase-3 genes (p <0.01) More obviously, its mechanism may be related to the regulation of bax, caspase-3 gene expression.
