目的采用RP-HPLC测定玄参配方颗粒中哈巴俄苷的含量。方法色谱柱为Hangbang C18(200mm×4.6mm,5μm),流动相为乙腈-1%醋酸溶液(27:73),检测波长为278nm,流速为1.0mL·min-1,以外标法峰面积定量。结果哈巴俄苷的线性范围为7.62～68.58μg·mL-1(r=0.9999),平均回收率为98.6%,RSD=1.65%。结论所建方法简便、快速、准确。
Objective To determine the content of harpagoside in Radix Scrophulariae by RP-HPLC. Methods The column was Hangbang C18(200mm×4.6mm, 5μm). The mobile phase was acetonitrile-1% acetic acid solution (27:73). The detection wavelength was 278nm. The flow rate was 1.0mL·min-1. . Results The linear range of harpagoside was 7.62-68.58 μg·mL-1 (r=0.9999). The average recovery was 98.6% and RSD was 1.65%. Conclusion The method is simple, rapid and accurate.
