目的观察30日龄FMR1基因敲除小鼠的旷场行为进行。方法在旷场实验中采取4个区域分割采用VIEWER软件及目测对30日龄的FMR1基因敲除小鼠和野生型小鼠进行旷场行为观察,数据采用多因素方差分析处理。结果采用VIEWER软件分析发现:FMR1基因敲除小鼠在第4区域的停留时间、运动长度、进入格次数、伸头较野生型小鼠显著增加,而在在第1区域的Tailmoves较野生型小鼠显著减少,FMR1基因敲除小鼠在各区的速度,站立不动与野生型小鼠无明显差别。FMR1基因敲除小鼠在第3区域的运动长度和进入格次数也较野生型小鼠显著增加,FMR1基因敲除小鼠在第2区域和第3区域的点头较野生型小鼠显著增加,P<0.05。采用目测分析发现:FMR1基因敲除小鼠在第4区域进入次数较野生型小鼠增加(P<0.05),FMR1基因敲除小鼠跨线次数较野生型小鼠显著增加,P<0.05。结论30日龄FMR1基因敲除小鼠的行为异常,运动性和兴奋性较野生型小鼠增高。采用VIEWER软件较目测更更加灵敏、客观、准确。
Objective To investigate the open field behavior of FMR1 knockout mice at 30 days of age. Methods Four open-field experiments were performed in the open-field experiment using VIEWER software and visual inspection of 30-day-old FMR1 knockout mice and wild-type mice under open-field behavior. The data were analyzed by multivariate analysis of variance. RESULTS: The results of VIEWER software showed that the FMR1 knockout mice had significantly increased residence time, length of movement and number of loci in the fourth area compared with the wild type mice, whereas the Tailmoves in the first area were smaller than the wild type Rats significantly reduced, FMR1 knockout mice in each region of the speed, stand still and wild-type mice no significant difference. FMR1 knockout mice in the third region of the movement length and the number of cells into the wild-type mice also significantly increased, FMR1 knockout mice in the second and third regions of nodding than wild-type mice increased significantly, P <0.05. Visual inspection showed that the number of FMR1 knockout mice in the fourth region was significantly higher than that in the wild type mice (P <0.05), and the number of crosses between the FMR1 knockout mice and the wild type mice was significantly increased (P <0.05). Conclusion The behavior, activity and excitability of 30-day-old FMR1 knockout mice are higher than those of wild type mice. Using VIEWER software more visually more sensitive, objective and accurate.
