目的 :探讨c-raf- 1反义寡脱氧核苷酸 (ASODN)对人卵巢癌裸鼠皮下移植瘤的治疗作用。方法 :将 15只裸鼠建立人卵巢癌裸鼠皮下移植瘤模型 ,然后随机分成 3组予以不同条件处理 :对照组 (腹腔注射转染液和脂质体 ) ,正义治疗组 (腹腔注射脂质体 -c -raf- 1-SODN) ,反义治疗组 (腹腔注射脂质体 -c -raf- 1-ASODN)。治疗期间定期观测裸鼠体重并测量肿瘤体积 ,计算抑瘤率及肿瘤缩小率。结果 :反义治疗组与正义治疗组的抑瘤率分别为 6 8.1%和 6 .8% ,反义治疗组肿瘤缩小率为 16 .7% ,与正义治疗组相比较有明显的差别。各组裸鼠体重变化无明显差别。结论 :脂质体 -c -raf - 1-ASODN对人卵巢癌裸鼠皮下移植瘤有一定的治疗价值 ,可能成为日后基因治疗的重要方向
Objective: To investigate the therapeutic effect of c-raf-1 antisense oligodeoxynucleotide (ASODN) on subcutaneous xenografts of human ovarian cancer in nude mice. METHODS: Fifteen nude mice were established subcutaneously xenografted model of human ovarian cancer in nude mice and then randomly divided into three groups for different conditions: control group (intraperitoneal injection of transfection liquid and liposome), the treatment group (intraperitoneal injection of lipids C-raf-1-SODN), antisense treatment group (intraperitoneal injection of liposome-c-raf-1-ASODN). During treatment, the body weight of nude mice was observed regularly and the tumor volume was measured. The tumor inhibition rate and tumor shrinkage rate were calculated. Results: The antitumor rates of the antisense treatment group and the positive treatment group were respectively 6.11% and 6.8%, while those of the antisense treatment group were 16.7%, which were significantly different from those of the control group. The body weight of nude mice in each group had no significant difference. Conclusion: Liposome-c -raf - 1 - ASODN has certain therapeutic value on subcutaneous xenografts of human ovarian cancer in nude mice and may be an important direction for future gene therapy
