目的:探讨瑞舒伐他汀预处理对大鼠脑缺血/再灌注损伤脑组织Bcl-2和Bax表达的影响。方法:48只SD大鼠随机分为假手术组(Sham组)、脑缺血/再灌注组(I/R组)、溶剂对照组(Vehicle组)和瑞舒伐他汀预处理组(Ros组)。Ros组在模型制备前用瑞舒伐他汀5 mg/kg/d连续灌胃10 d,1次/d;Vehicle组用相同体积的生理盐水连续灌胃10 d。线栓法制作大鼠大脑中动脉脑缺血/再灌注模型,于缺血2 h后再灌注24 h后行神经功能评分,断头取脑,免疫组织化学法和RT-PCR法观察脑组织Bcl-2和Bax表达水平的变化。结果:大鼠脑缺血/再灌注损伤后,Bcl-2和Bax显著增多;与I/R组和Vehicle组相比,Ros组神经功能改善,Bcl-2的表达显著上调,Bax的表达显著下调,Bcl-2/Bax比值显著升高,差异均有统计学意义(P<0.05)。结论:瑞舒伐他汀预处理对大鼠脑缺血/再灌注损伤有神经保护作用,其机制可能与上调脑组织中Bcl-2的表达,下调Bax的表达,Bcl-2/Bax比值增加,从而抑制胞凋亡有关。
Objective: To investigate the effect of rosuvastatin preconditioning on the expression of Bcl-2 and Bax in brain tissue after cerebral ischemia / reperfusion injury in rats. Methods: Forty eight SD rats were randomly divided into Sham group, I / R group, vehicle control group and Rosuvastatin pretreatment group (Ros group) ). Ros group was given rosuvastatin 5 mg / kg / d for 10 d once a day before model preparation. Vehicle group was given gavage for 10 d with the same volume of saline. The middle cerebral artery occlusion (MCAO) model was made by the method of thread occlusion. The neurological function scores, brain decapitation were taken 24 h after ischemia and reperfusion. Immunohistochemistry and RT-PCR were used to observe the changes of brain tissue Bcl-2 and Bax expression changes. Results: Compared with I / R group and Vehicle group, the neurological function, the expression of Bcl-2 and the expression of Bax in Ros group were significantly increased after cerebral ischemia / reperfusion injury in rats The ratio of Bcl-2 / Bax was significantly increased, the differences were statistically significant (P <0.05). Conclusion: Pretreatment with rosuvastatin has neuroprotective effects on cerebral ischemia / reperfusion injury in rats. The mechanism may be related to upregulation of Bcl-2 expression, down-regulation of Bax, increase of Bcl-2 / Bax ratio, Thus inhibiting apoptosis related.
