目的 :建立反相高效液相色谱法测定血浆中醋酸诺美孕酮的浓度。方法 :血浆样品用乙醚提取 ,选用WatersμBondapakC18(30 0mm× 3 9mm ,10 μm)为分析柱 ,甲醇 -水 (70∶30 )为流动相 ,流速为 1 2mL·min-1,米非司酮为内标 ,检测波长为 30 0nm。结果 :醋酸诺美孕酮线性范围为 0 1~ 10 μg·mL-1,最低检测浓度为 6 0ng·mL-1(S/N =3)。样品平均方法回收率为 95 2 0 % ,日内、日间精密度的RSD均小于 5 %。结论 :此方法稳定、可靠 ,可用于醋酸诺美孕酮血药浓度分析及药代动力学研究。
Objective: To establish a reversed-phase high performance liquid chromatography (HPLC) method for the determination of nomethorrel acetate in plasma. METHODS: The plasma samples were extracted with diethyl ether. WatersμBondapak C18 (30 0 × 39 mm, 10 μm) was used as the analytical column. Methanol-water (70:30) was used as the mobile phase at a flow rate of 12 mL · min-1. Mifepristone Internal standard, detection wavelength of 30 0nm. Results: The linear range of nomegestine acetate was 0 1 ~ 10 μg · mL-1 with the lowest detection limit of 60ng · mL-1 (S / N = 3). The average recovery rate of samples was 95 20%. The RSD of intra-day and inter-day precision was less than 5%. Conclusion: This method is stable and reliable, and can be used for the analysis of norephedrin acetate plasma concentration and pharmacokinetics.
