目的:优选抗柯萨奇B病毒性心肌炎胶囊醇提物大孔树脂纯化工艺。方法:以总皂苷、总黄酮和总木脂素含量为指标,考察树脂的吸附和解吸能力,筛选大孔树脂型号,采用单因素试验对最大上样量、上样流速、洗脱溶剂等进行考察。结果:非极性D101型大孔树脂效果最佳,优选的纯化工艺条件为醇提液生药质量浓度0.5 g.mL-1,药材-湿树脂量1∶4,吸附流速2 BV.h-1,3 BV水洗除杂,4 BV 70%乙醇洗脱。在此工艺条件下,总皂苷、总黄酮和总木脂素纯度分别由5.36%,0.3%,2.26%提高到53.82%,3.01%,22.87%。结论:D101型大孔树脂纯化抗柯萨奇B病毒性心肌炎胶囊醇提物,方法简便可行,纯化效果好,可推广使用。
Objective: To optimize the anti-Coxsackie B virus myocardium alcohol extract macroporous resin purification process. Methods: The total saponin, total flavonoids and total lignans content as an indicator to study the resin adsorption and desorption capacity, selection of macroporous resin models, single factor experiments on the maximum sample volume, the sample flow rate, elution solvent and so on Inspected Results: The non-polar D101 macroporous resin had the best effect. The optimal purification conditions were as follows: the crude drug concentration of 0.5 g.mL-1, the ratio of medicinal material to wet resin of 1: 4, the adsorption flow rate of 2 BV.h-1 , 3 BV water wash impurity, 4 BV 70% ethanol. Under these conditions, the purity of total saponin, total flavonoids and total lignans increased from 5.36%, 0.3% and 2.26% to 53.82%, 3.01% and 22.87%, respectively. Conclusion: D101 macroporous resin purified anti-Coxsackie B virus myocarditis alcohol extract, the method is simple and feasible, the purification effect is good, and can be widely used.
