目的建立土白术药材中(6E,12E)-十四癸二烯-8,10-二炔-1,3-二乙酸酯(TDEYA)和芹烷二烯酮两种化学成分的定性定量方法。方法采用薄层色谱法和高效液相色谱法。薄层色谱采用G254薄层硅胶预制板,以石油醚-乙酸乙酯(10∶1)为展开剂,置紫外光(254 nm)下检视。高效液相采用ZORBAX SB-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-1%甲酸溶液(67∶33),检测波长为254 nm,流速1.0 ml/min;柱温30℃;进样量10μl。结果 TDEYA和芹烷二烯酮在薄层色谱和高效液相色谱上均获得良好的分离,可作为土白术药材的定性定量指标。结论本法操作简单,结果准确,重现性好,可为评价土白术药材的质量标准提供检测方法。
Objective To establish a qualitative and quantitative method for the chemical constituents of (6E, 12E) -tetradecadien-8,10-diyne-1,3-diacetate (TDEYA) . Methods Thin layer chromatography and high performance liquid chromatography were used. Thin-layer chromatography using G254 thin-layer silica gel prefabricated plate, with petroleum ether - ethyl acetate (10: 1) as developing agent, under UV light (254 nm) under the view. The mobile phase was a methanol-1% formic acid solution (67:33) with a detection wavelength of 254 nm and a flow rate of 1.0 ml / min on a ZORBAX SB-C 18 column (250 mm × 4.6 mm, 5 μm) Column temperature 30 ℃; injection volume 10μl. Results TDEYA and cetadienone obtained good separation on TLC and HPLC and could be used as qualitative and quantitative indexes of Atractylodes. Conclusion This method is simple, accurate and reproducible. It can provide a method for the evaluation of the quality standards of medicinal herbs.
