目的:探讨稽留流产患者缺氧诱导因子(HIF-1α)与血管内皮生长因子(VEGF)在其血清和绒毛中的表达情况并进行二者相关性研究。方法:采用ELISA试验和SP法分别测定66例稽留流产患者(流产组)与54例正常早孕孕妇(对照组)血清和绒毛中HIF-1α和VEGF的表达水平,并将稽留流产组按有无明确原因分为有因组和无因组;同时根据胚胎死亡后稽留宫内时间分为>4周组、2~4周组和<2周组分析相关性。结果:流产组血清HIF-1α和VEGF的水平分别为(317.3±116.8)ng/L和(34.2±4.9)ng/L,对照组分别为(416.8±121.1)ng/L和(51.4±55.2)ng/L,两组差异有统计学意义(P<0.05);流产组患者绒毛中HIF-1α和VEGF的表达量分别为(3.1±2.2)ng/L和(3.7±2.1)ng/L,对照组分别为6.1±4.2和6.9±3.4,两组差异有统计学意义(P<0.05)。两组血清VEGF和HIF-1α的表达成呈正相关(P<0.05),两组绒毛中VEGF与HIF-1α表达呈正相关(P<0.05);但稽留宫内时间<2周组、2~4周组和>4周组血清和绒毛中HIF-1α和VEGF的表达比较差异均无统计学意义(P>0.05)。结论:HIF-1α和VEGF在血清和绒毛中的低表达可能是导致稽留流产的重要原因之一。
Objective: To investigate the expression of hypoxia-inducible factor (HIF-1α) and vascular endothelial growth factor (VEGF) in serum and villus of patients with missed abortion and to study their correlation. Methods: The expression of HIF-1α and VEGF in serum and villus of 66 cases of missed abortion (abortion group) and 54 cases of normal early pregnancy (control group) were determined by ELISA and SP method. The cause of the disease was clearly divided into causative and non-causative groups. At the same time, according to the intrauterine time after embryo death, it was divided into> 4 weeks group, 2 to 4 weeks group and <2 weeks group to analyze the correlation. Results: The levels of serum HIF-1α and VEGF in the abortion group were (317.3 ± 116.8) ng / L and (34.2 ± 4.9) ng / L respectively, while those in the control group were (416.8 ± 121.1) ng / L and (51.4 ± 55.2) ng / L, the difference between the two groups was statistically significant (P <0.05). The expression of HIF-1α and VEGF in villus of abortion group were (3.1 ± 2.2) ng / L and (3.7 ± 2.1) ng / L, The control group were 6.1 ± 4.2 and 6.9 ± 3.4 respectively, with significant difference between the two groups (P <0.05). There was a positive correlation between the expression of VEGF and HIF-1alpha in both groups (P <0.05). The expression of VEGF and HIF-1alpha in the two groups had a positive correlation (P <0.05) There was no significant difference in the expression of HIF-1α and VEGF in the serum and in the villus between weeks and> 4 weeks (P> 0.05). Conclusion: The low expression of HIF-1α and VEGF in serum and villi may be one of the important causes of missed abortion.
