目的探讨纺锤体组装检查点蛋白Mad1在原发性胃肠淋巴瘤(PGIL)中的表达及意义。方法选取2004年1月至2010年12月间武汉大学中南医院收治的21例原发性胃肠淋巴瘤患者的手术切除或者内镜活组织标本,同时收取患者的10份正常胃肠组织、8份反应性增生淋巴结和15份结内非霍奇金淋巴瘤组织为对照。采用免疫组化法检测Mad1、Ki-67的表达及幽门螺杆菌(Hp)感染,采用荧光原位杂交(FISH)方法检测Bcl-6基因易位情况。结果 Mad1在原发性胃肠淋巴瘤中的表达,高于正常胃肠组织和反应性增生淋巴结,差异均有统计学意义(均P<0.05),与结内非霍奇金淋巴瘤表达差异无统计学意义(P>0.05),Mad1表达与Ki-67表达呈正相关,差异有统计学意义(P<0.05)。与Bcl-6基因易位、Hp感染、性别、年龄、临床分期及国际预后指数等无关,差异均无统计学意义(均P>0.05)。结论 Mad1在PGIL中表达的增高与Ki-67增殖指数呈正相关,可成为对PGIL进行诊断和评估临床预后的生物学标志。
Objective To investigate the expression of spindle checkpoint protein Mad1 in primary gastrointestinal lymphoma (PGIL) and its significance. Methods Twenty-one patients with primary gastrointestinal lymphoma who underwent surgical resection or endoscopic biopsy from January 2004 to December 2010 in Central South Hospital of Wuhan University were enrolled. Ten normal gastric tissues A reactive hyperplastic lymph node and 15 non-Hodgkin’s lymphoma tissue as a control. The expression of Mad1 and Ki-67 and the infection of Helicobacter pylori (Hp) were detected by immunohistochemistry. The translocation of Bcl-6 gene was detected by fluorescence in situ hybridization (FISH). Results The expression of Mad1 in primary gastrointestinal lymphoma was significantly higher than that in normal gastrointestinal and reactive hyperplastic lymph nodes (all P <0.05), but not significantly different from those in non-Hodgkin’s lymphoma There was no statistical significance (P> 0.05). There was a positive correlation between Mad1 expression and Ki-67 expression (P <0.05). There was no significant difference between Bcl-6 gene translocation, Hp infection, sex, age, clinical stage and international prognosis index (all P> 0.05). Conclusion The increased expression of Mad1 in PGIL is positively correlated with Ki-67 proliferation index, which may be a biological marker for PGIL diagnosis and evaluation of clinical prognosis.
