目的对18α-甘草次酸(18α-Glycyrrhetinic acid,18α-GA)的制备工艺进行优化并结构表征。方法以天然甘草三萜皂苷元18β-甘草次酸(18β-GA)为原料,分别利用分步反应法和一锅法制得其光学异构体18α-GA,对2种制备方法的工艺和产率进行考察;用常规的四大光谱法确定化学结构。结果用分步反应法制得目标化合物的总产率为45%;而用一锅法可直接制得目标产物,产率达63%,简化了操作流程,缩短了反应时间,提高了反应产率;详细描述了目标化合物的光谱特征和理化性质。结论用一锅法制备18α-甘草次酸的操作简单,产率高,是制备光学纯度较高的18α-甘草次酸的较好途径。
Objective To optimize and characterize the preparation of 18α-Glycyrrhetinic acid (18α-GA). Methods The natural 18α-glycyrrhetinic acid (18β-GA) was used as raw material. The optical isomer 18α-GA was prepared by a one-pot method and a one-pot reaction. Rate of investigation; using the conventional four spectroscopy to determine the chemical structure. Results The total yield of the target compound was 45% by the step-reaction method. The yield of the target product was 63% by one-pot method, which simplified the operation procedure, shortened the reaction time and increased the reaction yield The spectral characteristics and physicochemical properties of the target compounds were described in detail. Conclusion The one-pot method for preparing 18α-glycyrrhetinic acid is simple and has a high yield. It is a good way to prepare 18α-glycyrrhetinic acid with high optical purity.
