目的 :研究屏氧酶 192谷氨酸 /精氨酸 (PON192 ,Gln/Arg)基因多态性与血浆脂质水平及心肌梗死(MI)的关系。方法 :应用多聚酶链反应 限制性内切酶片段长度多态性 (PCR RFLP)技术检测 132例MI与相同例数正常人PON 192基因多态性 ,血浆脂质水平测定按常规进行。结果 :发现PON 192基因型有三种 ,即纯合子突变型 (BB) ,杂合子突变型 (AB) ,正常型 (AA)。正常对照组中AA型频率 17.6 % ,AB型频率 4 5 .3% ,BB型频率37.1%。A等位基因频率为 4 5 .2 % ,B等位基因频率为 5 4 .8%。MI组AA型频率 10 .2 % ,AB型频率 4 1.3% ,BB型频率 4 8.5 %。A等位基因频率为 30 .7% ,B等位基因频率为 6 9.3%。两组对比等位基因频率差异有显著性意义。高密度脂蛋白水平在两组间差异无显著性意义。结论 :MI患者PON 192 ,B等位基因频率明显高于正常对照组 ,说明PON 192基因突变与MI的发生密切相关
Objective: To investigate the relationship between the polymorphisms of PON192, Gln / Arg and plasma lipid levels and myocardial infarction (MI). Methods: Polymerase chain reaction restriction fragment length polymorphism (PCR RFLP) was used to detect the polymorphism of PON 192 gene in 132 patients with MI and the same number of normal subjects. The plasma lipid levels were determined routinely. Results: There were three PON 192 genotypes, namely homozygous mutant (BB), heterozygous mutant (AB) and normal type (AA). In normal control group, AA frequency was 17.6%, AB frequency was 45.3%, BB frequency was 37.1%. The frequency of allele A was 45.2% and the frequency of allele B was 54.4%. In MI group, the frequency of AA type was 10.2%, the frequency of AB type was 4.3% and the frequency of BB type was 8.5%. A allele frequency was 30.7%, B allele frequency was 6 9.3%. There was significant difference between the two groups in allele frequency. High-density lipoprotein levels in the two groups showed no significant difference. Conclusion: The frequencies of PON 192 and B alleles in patients with MI were significantly higher than those in controls, indicating that the mutation of PON 192 is closely related to the occurrence of MI
