芘荧光发射光谱的第一振动带和第三振动带强度比值(I1/I3)能够反映周围微环境的极性水平,并且芘荧光光谱不受胶原蛋白内源荧光的干扰,作者用芘荧光探针考察了狭鳕鱼皮胶原蛋白在pH=7.2的磷酸缓冲液中的聚集行为。结果表明:当胶原蛋白质量浓度增大到一定程度时,芘荧光探针的I1/I3值有一突变,继续增大其质量浓度,I1/I3值趋向于定值,证明胶原蛋白在pH=7.2的磷酸缓冲液中,随质量浓度增大有明显的聚集行为。运用芘I1/I3值与胶原蛋白质量浓度对数的关系,通过作图法确定胶原蛋白的临界聚集质量浓度(CAMC)为0.138 g/L和0.48 g/L,结合不同质量浓度胶原蛋白溶液的聚集动力学曲线,故胶原蛋白的CAMC为0.48 g/L。由于胶原蛋白聚集过程在动力学上分为两个阶段,作者认为该胶原蛋白的聚集机理可能为协同成核生长机理。
