目的探讨辛伐他汀对人甲状腺未分化癌(ATC)细胞株HTh-74生长抑制和凋亡的作用。方法 MTT法检测细胞生长率;结晶紫染色观察细胞克隆形成情况;膜联蛋白Ⅴ/碘化丙啶(AnnexinⅤ/PI)双染检测细胞凋亡率;Western blot检测凋亡相关蛋白半胱氨酸蛋白酶3(Caspase-3)、B细胞淋巴瘤-白血病2(Bcl-2)和Bcl相关X蛋白(Bax)的表达。结果辛伐他汀对HTh-74细胞有显著生长抑制作用,呈浓度和时间依赖性(P<0.05或P<0.01)。辛伐他汀处理后可抑制细胞克隆形成、使细胞凋亡率显著增高、Caspase-3剪切体和Bax蛋白表达量增加、Bcl-2蛋白表达量降低、Bax/Bcl-2比值增加,且均呈浓度依赖性(P<0.05或P<0.01)。结论辛伐他汀可诱导人ATC细胞株HTh-74凋亡,其可能机制是与Bcl-2表达下调、Bax表达上调以及Caspase-3活化有关。
Objective To investigate the effect of simvastatin on the growth inhibition and apoptosis of human thyroid undifferentiated carcinoma cell line HTh-74. Methods MTT assay was used to detect the cell growth rate. The formation of cell clone was observed by crystal violet staining. The apoptosis rate was detected by AnnexinⅤ / PI double staining. The apoptosis related protein cysteine Caspase-3, B-cell lymphoma-leukemia 2 (Bcl-2) and Bcl-related protein X (Bax). Results Simvastatin had a significant inhibitory effect on HTh-74 cells in a concentration- and time-dependent manner (P <0.05 or P <0.01). After simvastatin treatment, cell cloning was inhibited, apoptosis rate was significantly increased, Caspase-3 splicing and Bax protein expression increased, Bcl-2 protein expression decreased, Bax / Bcl-2 ratio increased In a concentration-dependent manner (P <0.05 or P <0.01). Conclusion Simvastatin can induce the apoptosis of human ATC cell line HTh-74, which may be related to the down-regulation of Bcl-2 expression, the up-regulation of Bax expression and the activation of Caspase-3.
