干细胞因子(SCF)参与机体生长发育,调控多种细胞的生长,对造血干细胞、前B细胞、红系,粒系及巨核系细胞都有促分化作用,本试验主要探讨SCF对脐血NK前体细胞的促分化作用.本实验利用SCF,IL-2和SCF+IL-2培养脐血单个核细胞21d,动态计数单个核细胞总数,根据比例计算NK含量;并在第1,3,5,9,14和21天利用FACS检测细胞表型CD56/CD16和CD3分子;同时利用ATT法检测第1,5,9,14,和21天NK细胞的杀伤活性.实验结果显示:①SCF促进人脐血NK细胞的产生:SCF,IL-2和SCF+IL-2组中脐血单个核细胞的增殖倍数为3.5,4.1和6.8倍,根据脐血中NK细胞的含量为15%~20%,推算NK细胞的增殖倍数分别为5.3~7.0,6.15~8.2和10.2~13.6倍,而阴性对照组为2.5倍.由此可见,单独SCF不仅可以增加脐血NK细胞的绝对数量和比例,并且能够提高NK细胞对IL-2的增殖反应.②
