消栓颗粒对缺氧缺血性新生大鼠脑组织Caspase-3蛋白的影响

目的探讨消栓颗粒对缺氧缺血性脑损伤(HIBD)新生大鼠Caspase-3蛋白的影响。方法 7日龄SD新生大鼠36只,随机分为假手术组、HIBD模型组和消栓颗粒治疗组,每组各12只。HIBD模型组和消栓治疗组参照Yager法建立HIBD模型,消栓颗粒治疗组灌服消栓颗粒(8g/kg,qd),HIBD模型组及假手术组灌服等量9g/L氯化钠注射液。造模后第14d断头取出脑组织,观察3组大鼠左侧大脑半球病理形态学的变化,比较3组的病理学评分;免疫组化法检测3组大鼠海马及皮质区Caspase-3蛋白的表达情况。结果 HE染色结果显示,假手术组未见明显病变,HIBD模型组大鼠脑组织变性、坏死显著,消栓颗粒治疗组仅表现为局灶性神经细胞变性,病理学评分假手术组显著低于HIBD模型组及消栓颗粒治疗组(P<0.01),消栓颗粒组低于HIBD模型组(P<0.01)。免疫组化结果显示,假手术组海马及皮质区有少量Caspase-3蛋白表达,HIBD模型组Caspase-3蛋白的表达较假手术组明显增多,消栓颗粒治疗组Caspase-3蛋白的表达较HIBD模型组明显减少,3组间两两比较均具有显著性差异(P<0.01)。结论消栓颗粒可减轻HIBD新生大鼠的脑损伤,机制可能为抑制Caspase-3蛋白的表达。 Objective To investigate the effect of Xiaoshang Granule on Caspase-3 protein in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods Thirty-six SD neonatal rats, 7 days old, were randomly divided into sham-operated group, HIBD model group and Xiaoxiao Granule-treated group, with 12 rats in each group. The HIBD model group and the Hypoxia treatment group were treated with the Yager method to establish the model of HIBD. The HIBD model group and the sham operation group were fed with the same dosage of 9g / L NaCl (8g / kg, qd) Injection. The rats were sacrificed on the 14th day after decapitation to observe the histopathological changes of the left hemisphere in the three groups. The pathological scores of the three groups were compared. The expression of Caspase-3 in the hippocampus and cortex was detected by immunohistochemistry Protein expression. Results The results of HE staining showed that there was no obvious pathological changes in the sham operation group. The degeneration and necrosis of the brain tissue in the HIBD model group were significant. In the HXD group, the focal neuronal degeneration was only manifested. The pathological score was significantly lower in the sham operation group HIBD model group and Xiaoxiao Granule treatment group (P <0.01), Xiaoshi granules group was lower than HIBD model group (P <0.01). The results of immunohistochemistry showed that there was a small amount of Caspase-3 protein expression in the hippocampus and cortex in the sham operation group, Caspase-3 protein expression in the HIBD model group was significantly increased than that in the sham operation group, Caspase-3 protein expression The model group decreased significantly, and there was a significant difference between any two groups (P <0.01). Conclusion Xiao Shuan granules can reduce brain damage in HIBD neonatal rats, the mechanism may be inhibition of Caspase-3 protein expression.
