目的研究大鼠局灶性脑缺血-再灌注期细胞间粘附分子-1(ICAM-1)在转录水平表达规律。方法用插线法建立大鼠局灶性脑缺血再灌注模型,用逆转录-聚合酶链式反应(RT-PCR),测定ICAM-1转录水平(ICAM-1mRNA)的表达。结果发现ICAM-1mRNA在缺血2h升高,至缺血再灌注10h升至高峰,持续1周。结论ICAM-1在脑缺血再灌注时的表达明显上调,介导了白细胞、脑微血管内皮细胞的粘附增强,和脑缺血再灌注损伤关系密切。
Objective To investigate the expression of intercellular adhesion molecule-1 (ICAM-1) at focal cerebral ischemia-reperfusion in rats. Methods Focal cerebral ischemia-reperfusion model was established by the method of patch-line. The expression of ICAM-1 mRNA was determined by reverse transcription-polymerase chain reaction (RT-PCR) ICAM-1mRNA was found to be elevated at 2h after ischemia and peaked at 10h after ischemia-reperfusion for 1 week. Conclusion The expression of ICAM-1 is significantly up-regulated during cerebral ischemia-reperfusion, which leads to the enhanced adhesion of leukocytes and cerebral microvascular endothelial cells, which is closely related to cerebral ischemia-reperfusion injury.
