目的对新疆厚叶岩白菜素进行提取方法优化及不同部位含量的测定,为高效提取厚叶岩白菜素提供依据。方法利用正交实验,对醇提法的乙醇浓度、乙醇用量、提取时间、提取次数4个因素进行综合考虑、筛选,以及对厚叶岩白菜素不同部位进行含量测定。结果优选出最佳的乙醇提取新疆厚叶岩白菜素工艺条件为8倍量50%的乙醇,回流提取3次,每次0.5 h。厚叶岩白菜不同部位的岩白菜素差异具有统计学意义(P<0.05)。主根和须根中岩白菜素的含量较高,达到8.65%~9.58%,而叶中岩白菜素的含量相对较低,仅为0.15%。厚叶岩白菜素嫩叶含量显著高于老叶。结论本实验研究对高效提取新疆厚叶岩白菜素提供了有力的技术支撑,同时为新疆厚叶岩白菜药材质量评价奠定了理论基础。
OBJECTIVE To optimize the extraction method and the determination of the contents of the different parts of Brachycodon campestris in Xinjiang, and to provide the basis for the efficient extraction of Brachycodinium. Methods The orthogonal test was used to comprehensively consider and filter the four factors of ethanol concentration, ethanol consumption, extraction time and extraction times. Results The best ethanol extraction was obtained. The thick berberine in Xinjiang was extracted with 8 times the amount of 50% ethanol and refluxed three times for 0.5 h each. There were significant differences in the content of bergenin in different parts of Brachyceps kamoji (P <0.05). The main content of bergenin in the main roots and fibrous roots was 8.65% ~ 9.58%, while the content of bergenin in the leaves was relatively low, only 0.15%. The content of thick leaf cabbage leaves in young leaves was significantly higher than that of old leaves. Conclusion The experimental study provided a powerful technical support for the efficient extraction of thick-leaf bermudin in Xinjiang, and laid the theoretical foundation for the quality evaluation of the Chinese cabbage leaf-rich Chinese cabbage in Xinjiang.
