目的:探究灵芝乙醇提取物体内外抗肿瘤的活性。方法:体外实验:采用6种人肿瘤细胞系(胰腺癌SW1990、肺癌A549、宫颈癌Hela、肝癌Hep G2、胃癌MKN45、乳腺癌MCF-7),用灵芝乙醇提取物处理肿瘤细胞,观察细胞生长形态、绘制生长曲线、进行MTT实验及平板集落形成实验。体内实验:采用裸鼠胰腺癌SW1990移植瘤模型,以灵芝乙醇提取物50、100、150mg/kg剂量灌胃,计算瘤体积、抑瘤率及脏器系数。结果:体外实验:灵芝乙醇提取物对胰腺癌SW1990、肺癌A549、宫颈癌Hela、肝癌Hep G2、胃癌MKN45、乳腺癌MCF-7细胞的半数抑制率分别为96.62、44.21、78.66、117.60、44.34、73.28μg/ml。体内实验:70%灵芝提取物50、100、150mg/kg剂量组抑瘤率分别为37.2%、39.2%、43.9%。结论:体外实验表明灵芝乙醇提取物对上述6种肿瘤细胞均具有抑制增殖的作用,且浓度在40μg/ml以上时抑制增殖的作用较强。体内实验表明灵芝乙醇提取物能抑制胰腺癌SW1990生长。
Objective: To investigate the anti-tumor activity of ethanol extract of Ganoderma lucidum in vitro and in vivo. METHODS: In vitro experiments: Tumor cells were treated with six kinds of human tumor cell lines (pancreatic cancer SW1990, lung cancer A549, cervical cancer Hela, liver cancer Hep G2, gastric cancer MKN45, breast cancer MCF-7) Morphology, draw the growth curve, MTT experiments and plate colony formation experiments. In vivo experiments: The nude mice model of pancreatic cancer SW1990 xenografts was established by intragastric administration of Ganoderma lucidum ethanol extract at doses of 50, 100 and 150 mg / kg to calculate tumor volume, tumor inhibition rate and organ coefficient. Results: The in vitro experiments showed that the half inhibitory rates of Ganoderma lucidum ethanol extract to pancreatic cancer SW1990, lung cancer A549, cervical cancer Hela, liver cancer Hep G2, gastric cancer MKN45 and breast cancer MCF-7 were 96.62,44.21,78.66,117.60,44.34, 73.28 μg / ml. In vivo experiments: 70% Ganoderma lucidum extract 50,100,150 mg / kg dose group, the tumor inhibition rates were 37.2%, 39.2%, 43.9%. Conclusion: The in vitro experiments show that the extract of Ganoderma lucidum ethanol can inhibit the proliferation of above six kinds of tumor cells, and inhibit the proliferation when the concentration is above 40μg / ml. In vivo experiments show that Ganoderma lucidum ethanol extract can inhibit the growth of pancreatic cancer SW1990.
