目的:建立同时测定拈痛丸中盐酸小檗碱、盐酸巴马汀、盐酸药根碱、延胡索乙素含量的方法。方法:采用毛细管电泳法。电泳条件:未涂层弹性融硅石英毛细管柱(55 cm×75μm ID,有效长度47 cm);以100 mmol/L磷酸二氢钠溶液+50%乙醇(pH5.70)为运行缓冲液;分离电压21kV;重力进样5 s(高度15 cm);检测波长211 nm。结果:以盐酸麻黄碱为内标,盐酸小檗碱、盐酸巴马汀、盐酸药根碱、延胡索乙素的线性范围分别为(8.0~64.0)mg/mL(r=0.9989)、(2.5~12.5)mg/mL(r=0.9997)、(0.4~3.2)mg/mL(r=0.9982)、(0.2~1.6)mg/mL(r=0.9987)。结论:该法简便、快速、准确可靠,可用于拈痛丸的质量控制研究。
Objective: To establish a method for simultaneous determination of berberine hydrochloride, palmatine hydrochloride, jatrorrhizine hydrochloride and tetrahydropalmatine in Qutong Pill. Methods: Capillary electrophoresis. Electrophoresis conditions: uncoated elastic silica-silica capillary column (55 cm × 75 μm ID, effective length 47 cm); 100 mmol/L sodium dihydrogen phosphate solution + 50% ethanol (pH 5.70) as running buffer; separation Voltage 21 kV; gravity injection 5 s (height 15 cm); detection wavelength 211 nm. RESULTS: Using ephedrine hydrochloride as internal standard, the linear ranges of berberine hydrochloride, palmatine hydrochloride, jatrorrhizine hydrochloride and tetrahydropalmatine were respectively (8.0~64.0) mg/mL (r=0.9989), (2.5~ 12.5) mg/mL (r=0.9997), (0.4 to 3.2) mg/mL (r=0.9982), (0.2 to 1.6) mg/mL (r=0.9987). Conclusion: The method is simple, rapid, accurate and reliable, and can be used for the quality control of Qutong Pill.
