目的:观察骨钙素对小鼠3T3-L1前脂肪细胞分化的影响及对Sirt2、FoxO1、GLUT4和脂联素的影响,探讨骨钙素通过Sirt2/FoxO1通路控制肥胖、改善胰岛素抵抗、遏制2型糖尿病的机制。方法:将细胞接种到培养板中,加入诱导分化剂诱导其分化。用ELISA法检测各组中Sirt2、FoxO1以筛选最佳骨钙素剂量;油红O染色,观察细胞形态及脂肪含量变化,分光光度计510 nm波长处测量OD值。Real-Time PCR法检测各组中Sirt2、FoxO1、GLUT4和脂联素的表达。结果:骨钙素与Sirt2及FoxO1呈正相关(P<0.05)。A组细胞中Sirt2、FoxO1、GLUT4和脂联素的mRNA表达量均较B组升高(P<0.05)。C组细胞中Sirt2、FoxO1、GLUT4和脂联素的mRNA表达量较A组降低(P<0.05)。B、C组OD值明显高于A组(P<0.05)。结论:骨钙素可能通过Sirt2/FoxO1通路发挥作用调节脂联素及GLUT4的表达从而控制肥胖、改善胰岛素抵抗、可能有遏制2型糖尿病发生发展的作用。
OBJECTIVE: To observe the effect of osteocalcin on the differentiation of mouse 3T3-L1 preadipocytes and its effect on Sirt2, FoxO1, GLUT4 and adiponectin, and to explore the effect of osteocalcin on obesity and insulin resistance through the Sirt2 / FoxO1 pathway. Diabetes mellitus mechanism. Methods: The cells were inoculated into the culture plate, induced differentiation agent added to induce their differentiation. Sirt2 and FoxO1 in each group were detected by ELISA to select the best dose of osteocalcin. Oil red O staining was performed to observe the changes of cell morphology and fat content. OD value was measured at 510 nm wavelength of the spectrophotometer. Real-Time PCR was used to detect the expression of Sirt2, FoxO1, GLUT4 and adiponectin in each group. Results: Osteocalcin was positively correlated with Sirt2 and FoxO1 (P <0.05). The mRNA expression levels of Sirt2, FoxO1, GLUT4 and adiponectin in group A were significantly higher than those in group B (P <0.05). The mRNA expression of Sirt2, FoxO1, GLUT4 and adiponectin in group C was lower than that in group A (P <0.05). The OD values in group B and C were significantly higher than those in group A (P <0.05). CONCLUSION: Osteocalcin may play a role in regulating the expression of adiponectin and GLUT4 through Sirt2 / FoxO1 pathway to control obesity and improve insulin resistance. It may have the effect of inhibiting the development of type 2 diabetes.
