目的检测相关细胞因子血管生成素1(Ang1)、Ang2在大鼠局灶性脑缺血/再灌注后表达的动态变化。方法采用免疫组化及原位杂交技术。结果在正常脑组织及缺血/再灌注后均发现Ang1蛋白及Ang1mRNA的中等程度的广泛表达,在再灌注48h时达到高峰,此后虽有所下降,但仍为持续的阳性表达。Ang2蛋白及Ang2mRNA在对照组及假手术组中均呈阴性表达,在MCAO后6h于梗死灶内的一些单个细胞中发现,在12~24h后达到高峰,这种表达一直持续到72h。结论Ang1、Ang2在脑损伤的过程中起着重要的作用。
Objective To investigate the dynamic changes of angiopoietin 1 (Ang1) and Ang2 in rats after focal cerebral ischemia / reperfusion. Methods Immunohistochemistry and in situ hybridization were used. Results The normal expressions of Ang1 protein and Ang1 mRNA were found in normal brain tissue and after ischemia / reperfusion, and reached a peak at 48h after reperfusion. Afterwards, they remained persistent positive expression. Ang2 protein and Ang2 mRNA were negative in the control group and sham operation group, and were found in some single cells in the infarct zone at 6h after MCAO, reaching the peak after 12 ~ 24h, which lasted for 72h. Conclusion Ang1 and Ang2 play an important role in the process of brain injury.
