目的 探讨人前列腺干细胞的分离方法并初步鉴定其特性。方法 以器官捐献者的正常前列腺为研究对象 ,利用免疫磁珠细胞分离和胶原快速粘附方法 ,从前列腺基底细胞中分离前列腺干细胞 ,研究其克隆形成和分化特性。结果 从正常人前列腺内分离得到的细胞 ,能快速粘附胶原 ,克隆形成率明显高于未粘附的前列腺基底细胞 ,而且形成的克隆维持时间较长。结论 人前列腺基底细胞中存在极少量的具有高度增殖能力的一群细胞 ,它们具有干细胞的特征 ,但有待于进一步研究鉴定。
Objective To investigate the method of isolation of human prostatic stem cells and preliminary identification of its characteristics. Methods Normal prostate was collected from organ donors. Prostate stem cells were isolated from prostatic basal cells by immunomagnetic beads and rapid adhesion of collagen, and their clonogenicity and differentiation were studied. Results The cells isolated from the normal human prostate could rapidly adhere to the collagen. The colony formation rate was significantly higher than that of non-adherent prostate basal cells, and the formed colonies maintained for a long time. Conclusion There are a few cells with high proliferative capacity in human prostate basal cells, which have the characteristics of stem cells, but they need to be further studied and identified.
