目的探讨抑癌基因干扰素/维甲酸合并诱导的细胞凋亡基因(GRIM-19)在乳腺癌与良性乳腺疾病组织中表达的差异,及其与转录信号转导子及激活子3(STAT3)的相关性及其临床意义。方法运用免疫组化方法,研究64例乳腺癌组织(乳癌组)与30例良性乳腺疾病乳腺组织(对照组)中的GRIM-19和STAT-3蛋白的表达,以及两者的相关性。结果 GRIM-19蛋白在乳腺癌组织中的表达明显低于对照组,而STAT-3蛋白在乳癌组中的表达明显高于对照组,差别均有统计学意义(P<0.01)。GRIM-19蛋白在三阴乳腺癌中的表达显著高于非三阴乳腺癌,差别有统计学意义(P<0.05)。乳腺癌中GRIM-19蛋白与STAT-3蛋白的表达呈负相关(r=-0.524,P<0.01)。结论在乳腺癌组织中,同时存在GRIM-19的低表达和STAT-3的高表达,可能导致细胞的恶性转化,从而促进乳腺癌的发生发展。
OBJECTIVE: To investigate the difference of expression of tumor suppressor gene interferon / retinoic acid-induced apoptosis gene (GRIM-19) in breast cancer and benign breast disease and its relationship with transcription signal transducers and activator of transcription 3 (STAT3) Correlation and clinical significance. Methods The expressions of GRIM-19 and STAT-3 protein in 64 cases of breast cancer (breast cancer group) and 30 cases of benign breast disease (control group) were studied by immunohistochemistry. Results The expression of GRIM-19 protein in breast cancer was significantly lower than that in control group, while the expression of STAT-3 protein in breast cancer was significantly higher than that in control group (P <0.01). The expression of GRIM-19 protein in triple-negative breast cancer was significantly higher than that in non-triple-negative breast cancer (P <0.05). The expression of GRIM-19 protein and STAT-3 protein in breast cancer were negatively correlated (r = -0.524, P <0.01). Conclusions In breast cancer, both low expression of GRIM-19 and high expression of STAT-3 may lead to malignant transformation of cells and promote the development of breast cancer.
