目的探讨柴油车尾气颗粒物有机提取物(DEPE)对中国仓鼠肺成纤维细胞(V79)胞内游离Ca~(2+)[Ca~(2+)]_i的影响及其机制。方法采用Ca~(2+)指示剂Fura-2/AM荧光负载检测技术,研究DEPE对细胞内[Ca~(2+)]_i浓度的影响。结果在一定剂量范围内,随着DEPE染毒剂量增加或时间延长,细胞内[Ca~(2+)]_i浓度升高,与胞内钙库释放和胞外Ca~(2+)内流有关,并以胞外Ca~(2+)内流为主。结论DEPE能致细胞内[Ca~(2+)]_i升高,氧化损伤是导致细胞内[Ca~(2+)];升高的机制之一。
Objective To investigate the effect of diesel particulate matter (DEPE) on the intracellular Ca ~ (2 +) [Ca ~ (2 +)] _ i in Chinese hamster lung fibroblasts (V79) and its mechanism. Methods Fura-2 / AM fluorescent loading assay was used to study the effect of DEPE on [Ca 2+] i concentration in cells. Results Within a certain dosage range, with the increase of dose of DEPE or prolongation of time, the concentration of intracellular [Ca 2 +] i was increased, and the intracellular Ca 2+ release and extracellular Ca 2+ influx Related, and the main extracellular Ca ~ (2 +) influx. Conclusions DEPE can cause intracellular [Ca2 +] i increase, and oxidative damage is one of the mechanisms leading to [Ca2 +] increase in cells.
