VIM金属β-内酰胺酶是1种能水解几乎所有(除氨曲南等单环外)的β-内酰胺类抗生素,也是已报道蛋白质中与新德里金属β-内酰胺酶(NDM-1)序列相似性最高的一类获得性金属β-内酰胺酶。VIM-1与NDM-1相似,前者基因易于在细菌间传播和扩散,迄今还未发现临床有效的抑制剂,所以研发新型有效的抑制剂迫在眉睫。综述了近年来VIM序列、结构特点、耐药机制及相关抑制剂的最新研究进展,为设计、筛选、开发新型VIM-1抑制剂奠定基础,亦可为深入了解NDM-1设计开发其抑制剂提供帮助。
VIM metallo-β-lactamase is a β-lactam antibiotic that hydrolyzes almost all (except monosclers such as aztreonam) and also has been reported to bind to New Delhi metallo-β-lactamase (NDM-1) A class of acquired metal β-lactamases with highest sequence similarity. VIM-1 is similar to NDM-1. The former gene is easy to spread and spread among bacteria. So far, no clinically effective inhibitor has been found. Therefore, it is urgent to develop new and effective inhibitors. This review summarizes recent advances in VIM sequences, structural features, drug resistance mechanisms and related inhibitors and provides a basis for the design, screening and development of novel VIM-1 inhibitors. It may also provide insights into the design and development of inhibitors of NDM-1 provide help.
