目的探讨压力超负荷下,钙蛋白酶参与心肌肥厚的信号转导机制。方法16只SD大鼠随机均分为模型组和对照组。模型组采用不完全结扎腹主动脉法建立大鼠高血压心肌肥厚模型。放射免疫法检测两组心肌心房肽(ANF)、血管紧张素Ⅱ(AngⅡ)水平;Western blot法检测左室心肌钙蛋白酶的表达水平。结果模型组术后血压、心肌ANF、AngⅡ含量、左室心肌钙蛋白酶同工酶u表达水平明显高于对照组(P<0.01)。结论心肌肥厚组织中钙蛋白酶同工酶u表达增加,钙调神经磷酸酶信号通路参与了心肌肥厚的发病过程。
Objective To investigate the signal transduction mechanism of calpain involved in cardiac hypertrophy under pressure overload. Methods Sixteen SD rats were randomly divided into model group and control group. The model group was established by incomplete abdominal ligation of the rat model of hypertensive cardiac hypertrophy. The levels of myocardial atrial natriuretic peptide (ANF) and angiotensin Ⅱ (AngⅡ) were measured by radioimmunoassay. The expression of cardiac calpain was detected by Western blot. Results The postoperative blood pressure, myocardial ANF, Ang Ⅱ content and the expression of cardiac ventricular calpain isoenzyme u in the model group were significantly higher than those in the control group (P <0.01). Conclusions The expression of calpain isoenzyme u is increased in cardiac hypertrophy, and the calcineurin signaling pathway is involved in the pathogenesis of cardiac hypertrophy.
