目的研究HMGB1在葡萄胎、绒毛膜癌组织及正常早期妊娠绒毛中的表达及与葡萄胎发生的关系。方法选取2012年6月-2014年12月在该院妇产科门诊要求人工流产的35例为正常组;2008年6月-2014年12月在潍坊市中心医院、潍坊市人民医院、潍坊市妇幼保健院因葡萄胎、绒毛膜癌住院的为葡萄胎组和绒毛膜癌组,各20例。运用免疫组织化学方法检测HMGB1在绒毛组织中的定位,通过逆转录聚合酶链反应法检测绒毛膜癌、葡萄胎及正常早期妊娠绒毛组织中HMGB1的核酸表达水平,通过免疫印迹法检测HMGB1蛋白表达的水平。结果免疫组织化学技术显示,HMGB1主要表达于细胞质和细胞核,RT-PCR及免疫印迹结果显示,绒毛膜癌和葡萄胎组织HMGB1的表达量高于正常绒毛组织,差异有统计学意义(P<0.05)。结论与正常妊娠绒毛组织相比,绒毛膜癌和葡萄胎中HMGB1表达发生改变,提示HMGB1在葡萄胎和绒毛膜癌的发病机制中可能发挥了作用。
Objective To study the expression of HMGB1 in hydatidiform mole, choriocarcinoma and normal early gestational chorion and its relationship with hydatidiform mole. Methods From June 2012 to December 2014, 35 cases of induced abortion in obstetrics and gynecology clinic of the hospital were selected as normal group. From June 2008 to December 2014, we performed the operation in Weifang Central Hospital, Weifang People’s Hospital, Weifang City Maternal and Child Health Hospital due to hydatidiform mole, choriocarcinoma hospitalized for hydatidiform mole and choriocarcinoma group, 20 cases each. Immunohistochemistry was used to detect the localization of HMGB1 in villi. The expression of HMGB1 in choriocarcinoma, hydatidiform mole and normal early pregnancy villi was detected by reverse transcription-polymerase chain reaction (RT-PCR). The expression of HMGB1 protein was detected by Western blotting s level. Results Immunohistochemistry showed that HMGB1 was mainly expressed in the cytoplasm and nucleus. The results of RT-PCR and Western blotting showed that the expression of HMGB1 in choriocarcinoma and hydatidiform mole was higher than that in normal chorion (P <0.05 ). Conclusion The expression of HMGB1 in choriocarcinoma and hydatidiform mole is changed compared with that in normal pregnancy, suggesting that HMGB1 may play a role in the pathogenesis of hydatidiform mole and choriocarcinoma.
