目的探讨人类重组拉-巨噬细胞集落刺激因子(rhGM—CSF)对照射狗脾脏及淋巴结修复作用的影响.方法雄性Beagle狗14只.均双例髂骨屏蔽,其余部位用60Coγ射线照射.除对照组外.治疗组股静脉注射给药.连续20d,低剂量组为10μg·kg-1·d-1.高剂量组20μg·kg-1·d-1.30d时处死动物.用HE和AgNOR染色观察脾脏和淋巴结结构的变化,并对脾小体和淋巴滤泡面密度及数密度进行定量分析.结果低剂量组淋巴结和脾脏组织结构中等程度恢复;淋巴细胞增殖不十分明显.而高剂量组的牌及淋巴结基本恢复正常,淋巴细胞增殖明显高于对照组.结论rhGM—CSF可促进照射后构狗脾脏及淋巴结形态与功能的恢复,其恢复程度与该因子的剂量有关.
Objective To investigate the effect of human recombinant human macrophage colony-stimulating factor (rhGM-CSF) on spleen and lymph node repair in irradiated dogs. Methods Male Beagle dogs 14. Double cases of ilium shield, the rest of the 60Coγ-ray irradiation. In addition to the control group. The treatment group femoral vein injection. For 20 consecutive days, the low-dose group was 10 μg · kg-1 · d-1. The animals were sacrificed at high dose of 20μg · kg-1 · d-1.30d. The changes of spleen and lymph node structure were observed with HE and AgNOR staining, and the surface density and number density of spleen body and lymphoid follicle were quantitatively analyzed. Results Low-dose group of lymph node and spleen tissue structure restored to a certain extent; lymphocyte proliferation is not very clear. The high-dose group of cards and lymph nodes returned to normal, lymphocyte proliferation was significantly higher than the control group. Conclusion rhGM-CSF can promote the recovery of morphology and function of spleens and lymph nodes in dogs after irradiation, and the degree of recovery is related to the dosage of this factor.
