目的:探讨参附益心颗粒对慢性心力衰竭大鼠血浆与心肌血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)水平的影响。方法:随机将150只雄性SD大鼠分为造模组130只与假手术组20只,造模组结扎左冠状动脉前降支,假手术组仅冠状动脉前降支下穿线不结扎。造模8周后,经超声测定大鼠左室射血分数(left ventricular ejection fraction,LVEF)<50%者随机分为5组(卡托普利组、氯沙坦组、参附益心颗粒(5.5,21.9 g.kg-1)2个剂量组、模型组],每组10只,另选假手术组大鼠10只。假手术组及模型组以纯净水ig,药物干预组将药物溶于纯净水中ig给药,每天1次,4周后检测LVEF、腹主动脉采血、取心肌组织检测AngⅡ。结果:参附益心颗粒21.9 g.kg-1组有明显改善LVEF作用,参附益心颗粒2个剂量组均明显降低血浆及心肌AngⅡ水平,且作用与卡托普利相当。结论:参附益心颗粒可改善心衰大鼠心功能,下调血浆及心肌AngⅡ水平,阻断RAS,可能起到延缓或改善心肌重塑的作用。
Objective: To investigate the effect of Shenfu Yixin Granule on plasma and myocardial angiotensin Ⅱ (AngⅡ) in rats with chronic heart failure. Methods: One hundred and fifty male SD rats were randomly divided into model group (130 rats) and sham operation group (20 rats). Rats in model group were ligated with left anterior descending coronary artery. Sham operation group was ligated only without descending anterior descending coronary artery. After 8 weeks of modeling, rats with left ventricular ejection fraction (LVEF) <50% were randomly divided into 5 groups (captopril group, losartan group, Shenfu Yixin Granule (5.5,21.9 g.kg-1) 2-dose group, model group], 10 rats in each group and 10 rats in sham-operation group.The rats in sham operation group and model group were treated with pure water ig and drug intervention group Dissolved in purified water ig administration, once a day, 4 weeks after the detection of LVEF, abdominal aorta blood, myocardial tissue was taken to detect Ang Ⅱ.Results: Shen Fu Yi Xin particles 21.9 g.kg-1 group significantly improved the role of LVEF Both the two groups of Xixinxin Granule could significantly reduce AngⅡ level in plasma and myocardium, and the effect was similar to that of captopril.Conclusion: Shenfu Yixin Granule can improve heart function and decrease AngⅡ level in plasma and myocardium of rats with heart failure Off RAS, may play a role in delaying or improving myocardial remodeling.
