目的:观察黄药子醇提物对人胃癌细胞系裸鼠移植瘤的抑瘤作用及白细胞介素-8(IL-8)和可溶性细胞间黏附分子-1(sICAM-1)表达水平的影响。方法:建立胃癌裸鼠皮下移植瘤模型,造模成功后随机分为模型对照组,5-Fu治疗组,黄药子醇提物低、高剂量组,每组10只,分别采用皮下注射生理盐水、腹腔注射5-Fu(30mg/kg)、腹腔注射黄药子醇提物(50mg/kg和100mg/kg)进行治疗。治疗3周后处死动物,眼眶取血,酶联免疫吸附法(ELISA)检测血清IL-8和sICAM-1含量;剥取肿瘤,测量计算肿瘤体积并称取体质量,计算抑瘤率。结果:黄药子高剂量组瘤体积和体质量明显小于模型对照组(P<0.05),黄药子低、高剂量组之间瘤体积和体质量比较差异无统计学意义(P>0.05),5-Fu组和黄药子低、高剂量组三者对瘤体积的抑制率分别为25.00%、17.86%和39.29%,对瘤体质量的抑制率分别为37.50%、25.00%和43.75%。5-Fu组和黄药子低、高剂量组裸鼠血清IL-8和sICAM-1水平显著低于模型对照组(P<0.05);黄药子高剂量组血清IL-8和sI-CAM-1水平明显低于低剂量组(P<0.05);黄药子高剂量组血清sICAM-1水平明显低于5-Fu组(P<0.05)。结论:黄药子醇提物对裸鼠胃癌移植瘤模型有明显的抑瘤作用,其作用机制可能与下调细胞因子IL-8和sICAM-1的表达有关。
OBJECTIVE: To observe the antitumor effect of extracts of xanthium salsa on human gastric cancer cell line xenografts in nude mice and the expression of interleukin-8 (IL-8) and soluble intercellular adhesion molecule-1 (sICAM-1). Methods: A subcutaneous xenograft model of gastric cancer in nude mice was established. After the model was successfully established, the rats were randomly divided into model control group, 5-Fu treatment group and low and high dose of xanthium alcohol extract group, 10 rats in each group. Subcutaneous injection of normal saline, Intraperitoneal injection of 5-Fu (30mg / kg), intraperitoneal injection of xanthophyllol extract (50mg / kg and 100mg / kg) for treatment. After 3 weeks of treatment, animals were sacrificed and orbital blood was collected. Serum levels of IL-8 and sICAM-1 were detected by enzyme-linked immunosorbent assay (ELISA). Tumors were excised and tumor volume was measured and weighed to calculate tumor inhibition rate. Results: The tumor volume and body weight of the high-dose group were significantly lower than those of the model control group (P <0.05), while there was no significant difference in the tumor volume and body weight between the low and high dose groups (P> 0.05) The inhibitory rates of the three groups on the tumor volume were 25.00%, 17.86% and 39.29%, respectively, and the inhibition rates on the tumor mass were 37.50%, 25.00% and 43.75%, respectively. Serum levels of IL-8 and sICAM-1 in 5-Fu and xanthium low and high dose groups were significantly lower than those in model control group (P <0.05); serum IL-8 and sI-CAM-1 levels were significantly increased (P <0.05). The level of sICAM-1 in the high-dose group was significantly lower than that in the 5-Fu group (P <0.05). CONCLUSION: Alcohol extract of Radix Astragali can significantly inhibit tumor growth in nude mice with gastric cancer and its mechanism may be related to down-regulation of cytokines IL-8 and sICAM-1.
