本研究以华蟾素对喉癌细胞株Hep-2生长及癌基因的表达作为观测指标,研究中药对人喉癌细胞的作用,以探讨中西药结合对喉癌综合治疗的可能性.人喉癌Hep-2细胞株为北京耳鼻喉科研究所提供.华蟾素(cinobufacini-CIN),安徽淮北生化制药厂生产.p53,C-myc,单抗购自北京中山生物技术有限公司;bcl-2单抗购自DAKO公司.酶联免疫检测仪(国产).FACS440流式细胞分析仪(美国).观察指标:①形态学观察;②MTT试验;③p53,C-myc,bcl-2癌基因产物表达检测;④流式细胞仪检测分析.结果:(1)CIN对Hep-2细胞作用后细胞形态学改变:①光镜下观察:在倒置显微镜下可见对照组细胞贴壁生长.CIN试验组短时间组细胞增
In this study, Huachansu was used to observe the growth of laryngeal carcinoma cell line Hep-2 and the expression of oncogene as an observational index to study the effect of traditional Chinese medicine on human laryngeal carcinoma cells in order to explore the possibility of integrated treatment of laryngeal carcinoma with Chinese and Western medicine. The cancer Hep-2 cell line was provided by the Beijing Institute of Otolaryngology. Cinobufacini-CIN, produced by Anhui Huaibei Biochemical Pharmaceutical Factory. p53, C-myc, monoclonal antibody purchased from Beijing Zhongshan Biotechnology Co., Ltd.; bcl- 2 Monoclonal antibody purchased from DAKO Corporation. Enzyme-linked immunosorbent assay (domestic). FACS440 Flow Cytometry (USA). Observations: 1 Morphological observation; 2MTT assay; 3p53, C-myc, bcl-2 oncogene product Expression test; 4 Flow cytometry analysis. Results: (1) Morphological changes of CIN on Hep-2 cells: 1 Observed under light microscope: Adherent growth of control cells was observed under inverted microscope. CIN test group Short-term cell increase
