目的:检测新疆哈萨克族(哈族)及正常人群中低分子量蛋白酶体(LMP7)基因多态性的分布,探讨LMP7基因遗传多态性与新疆哈族食管鳞癌易感性的相关性。方法:应用PCR-RFLP方法检测168例哈族食管癌石蜡组织和283例同一地区无肿瘤病史的哈族正常人群血样LMP7基因型分布。应用SPSS11.5对实验结果进行统计学分析,并进行Hardy-Weinberg检验。结果:在新疆哈族食管癌组中,LMP7三种基因型Q/Q、Q/K、K/K所占比例分别是81.55%、14.88%、3.57%,与哈族正常对照组中68.55%、29.68%、1.77%相比差异有统计学意义(χ2=13.39、P<0.05),杂合型LMP7Q/K降低机体患食管癌风险0.42倍(OR=0.42 95%CI=0.26~0.70)。结论:LMP7基因遗传多态性与新疆哈族食管鳞癌易感性相关联;其中杂合型LMP7Q/K可能为新疆哈萨克族食管鳞癌患者的保护因素。
Objective: To detect the distribution of low molecular weight proteasome (LMP7) gene polymorphism in Xinjiang Kazak (Kazakh nationality) and normal population, and to explore the relationship between LMP7 genetic polymorphism and susceptibility to Kazakh esophageal squamous cell carcinoma in Xinjiang. Methods: PCR-RFLP was used to detect the distribution of LMP7 genotypes in 168 cases of Kazakh esophageal paraffin tissues and 283 normal Han nationality without cancer history. The results of SPSS11.5 were used for statistical analysis and Hardy-Weinberg test. Results: The proportions of Q / Q, Q / K and K / K of LMP7 genotypes were 81.55%, 14.88% and 3.57% respectively in Kazakh Ha esophageal cancer group and 68.55% in Kazakh normal control group , 29.68% and 1.77%, respectively (χ2 = 13.39, P <0.05). The heterozygous LMP7Q / K reduced the risk of esophageal cancer by 0.42 times (OR = 0.42 95% CI = 0.26-0.70). CONCLUSION: Genetic polymorphisms of LMP7 gene are associated with susceptibility to Kazakh esophageal squamous cell carcinoma in Xinjiang. Among them, heterozygous LMP7Q / K may be a protective factor for patients with esophageal squamous cell carcinoma in Xinjiang Kazakh.
