目的通过正交实验优选纤维素酶法提取匙羹藤茎总皂苷的最佳提取工艺。方法匙羹藤总皂苷在5%香草醛-冰醋酸和高氯酸的混合体系中显色,于500 nm处有最大吸收,用比色法测总皂苷含量,以总皂苷的产量为考察指标。采用酶解-乙醇浸提,正丁醇萃取,匙羹藤总皂苷提取率作为评价指标,通过单因素试验和正交试验,确定提取最佳工艺。结果酶解法提取匙羹藤总皂苷的最佳工艺为:酶解温度50℃,酶解时间3.0h,酶用量为8mg纤维素/g样品,p H5.5,固液比1∶10条件下,匙羹藤总皂苷提取率最高。结论此最佳工艺可用于匙羹藤茎总皂苷的提取。
OBJECTIVE To extract the best extraction technology of total saponin from Cunninghamia lanceolata through orthogonal experiment with cellulase. Methods Total giganoton saponin was developed in a mixed system of 5% vanillin-acetic acid and perchloric acid and showed the maximum absorption at 500 nm. The total saponin content was determined by colorimetry and the total saponin yield was taken as the index . The optimum extraction conditions were determined by enzymolysis - ethanol extraction, n - butanol extraction and the extraction rate of total saponin of Gymnema sibirica by single factor test and orthogonal experiment. Results The optimum conditions for enzymatic hydrolysis were as follows: enzymatic hydrolysis temperature 50 ℃, enzymolysis time 3.0h, enzyme dosage 8mg cellulose / g sample, p H 5.5, solid-to-liquid ratio 1:10 , Gymnemia total ginsenoside extraction rate. Conclusion This best technique can be used for the extraction of total saponin from Stem and Cane.
