目的:探讨以壳聚糖为佐剂的空肠弯曲菌基因疫苗免疫小鼠后的Th细胞免疫应答状态。方法:昆明小鼠随机分组:设空白对照组、空载体组、pcDNA3.1(-)-peb1A组、壳聚糖+pcDNA3.1(-)-peb1A组,各组均采用小鼠股四头肌注射法。在第0、10、20天免疫,于每次免疫后第10天采集各组小鼠血浆,间接ELISA法检测免疫后不同时间小鼠血浆中Th1、Th2细胞因子的含量。结果:Th1类细胞因子IL-2、IFN-γ,在第10、20、30天,裸DNA组、佐剂DNA组、空载体组及NS组两两相比均无显著性差异(P>0.05);Th2类细胞因子IL-4、IL-10:实验组高于两对照组,在IL-4的检测中有显著性差异(P<0.05),在IL-10的检测中裸DNA组高于空白对照组有显著性差异(P<0.05),其裸DNA组高于空载体组,仅在第20天有显著性差异(P<0.05);佐剂DNA组高于裸DNA组,在IL-4的检测中有显著性差异(P<0.05),在IL-10的检测中,仅在第10、20天有显著性差异(P<0.05);佐剂DNA组高于两对照组,有显著性差异(P<0.05)。结论:以壳聚糖为佐剂的空肠弯曲菌基因疫苗可促进Th2细胞为主的免疫反应。
Objective: To investigate the Th cell immune response in mice immunized with Campylobacter jejuni gene vaccine adjuvanted with chitosan. Methods: Kunming mice were randomly divided into control group, empty vector group, pcDNA3.1 (-) - peb1A group and chitosan + pcDNA3.1 (-) - peb1A group. Muscular injection. The mice were immunized on days 0, 10 and 20, and the plasma of mice in each group was collected on the 10th day after each immunization. The levels of Th1 and Th2 cytokines in the plasma of mice at different times after immunization were detected by indirect ELISA. Results: There was no significant difference in Th1 cytokines IL-2 and IFN-γ between the naked DNA group, adjuvant DNA group, empty vector group and NS group on the 10th, 20th and 30th day (P> 0.05). The levels of IL-4 and IL-10 in Th2 cytokines were higher in the experimental group than in the two control groups, and there was a significant difference (P <0.05) in the detection of IL-4. (P <0.05). Compared with the control group, the nude DNA group was higher than the blank group, the difference was significant only on the 20th day (P <0.05) There was a significant difference in the detection of IL-4 (P <0.05). There was a significant difference in the detection of IL-10 only on the 10th and 20th days (P <0.05) Group, there was significant difference (P <0.05). Conclusion: Chitosan adjuvanted Campylobacter jejuni gene vaccine can promote Th2 cell-based immune response.
