目的:观察铁皮石斛多糖对冠心病模型家兔心功能、心肌收缩能力的影响,并探讨其相关作用机制。方法:30只健康家兔随机分为对照组、模型组、铁皮石斛多糖低、中、高剂量组5组,每组6只。采用左冠状动脉放置缩窄环方法,制备冠心病模型。低、中、高剂量组铁皮石斛多糖按100,200,400 mg·kg-1ig给予,每天1次,连续21 d。对照组、模型组ig给予等体积生理盐水。记录左室收缩压(LVSP),左室舒张末压(LVEDP),左室压力变化最大上升和下降速率(±dp/dtmax),动脉收缩压(BPs)和动脉舒张压(BPd),心室质量指数,RT-PCR法检测心肌细胞α-肌球蛋白(α-MHC)mRNA,β-肌球蛋白(β-MHC)mRNA表达,免疫组化检测肌动蛋白(actin)表达。结果:与对照组比较,模型组LVEDP,LVWI,RVWI均升高(P<0.01,P<0.05),而LVSP,±dp/dtmax,BPs均降低(P<0.01,P<0.05),α-MHC mRNA表达显著下降(P<0.01),β-MHC mRNA升高(P<0.05),模型组actin蛋白表达呈阳性的心肌细胞较多。与模型组比较,铁皮石斛多糖低、中剂量组LVEDP降低(P<0.01,P<0.05),而LVSP,±dp/dtmax,BPs均升高(P<0.01,P<0.05),α-MHC mRNA表达显著升高(P<0.01),β-MHC mRNA表达下降(P<0.05),各剂量组actin蛋白表达呈阳性的心肌细胞较少。结论:铁皮石斛多糖一定程度上恢复心肌舒缩能力、改善心功能。
Objective: To observe the effect of Dendrobium candidum polysaccharide on heart function and myocardial contractility of coronary heart disease model rabbits and to explore its related mechanism. Methods: Thirty healthy rabbits were randomly divided into 5 groups: control group, model group and Dendrobium candidum polysaccharide at low, middle and high doses, with 6 in each group. Results: Compared with the control group, LVEDP, LVWI and RVWI in model group were significantly increased (P <0.01, P <0.05), LVSP, ± dp / dtmax and BPs were decreased The expression of MHC mRNA was significantly decreased (P <0.01) and β-MHC mRNA was increased (P <0.05). The expression of actin in the model group was more than that in the model group. Compared with the model group, the LVEDP of low and middle doses of Dendrobium candidum decreased (P <0.01, P <0.05), and the levels of LVSP, ± dp / dtmax and BPs increased (P <0.01), while the expression of β-MHC mRNA decreased (P <0.05). There was less cardiomyocytes with actin protein expression in each dose group. Conclusion: Dendrobium candidum polysaccharide can restore myocardial contractility to a certain extent and improve cardiac function.
