紫杉烷类药物主要包括紫杉醇、多西他赛,以及具有紫杉烷骨架结构的衍生物,它们具有高效广谱的抗肿瘤活性,然而制剂开发却一直受到其自身水溶性差的限制。纳米给药系统在实现靶向性给药、缓释给药、提高难溶性药物溶解度以及降低药物不良反应等方面均表现出良好的应用前景,因而采用纳米载体传递紫杉烷类药物得到广泛关注。综述了近几年紫杉烷类药物纳米制剂的研究进展,以期为新型药物剂型的开发和应用提供参考。
Taxanes include mainly paclitaxel, docetaxel, and derivatives with a taxane backbone structure that have broad spectrum of antitumor activity, however, formulation development has been limited by its poor water solubility. Nano-drug delivery system has shown a good application prospect in the aspects of targeted drug delivery, sustained release drug delivery, drug solubility enhancement and drug adverse reactions reduction, and thus widespread attention has been paid to the use of nano-carriers to deliver taxanes . In this paper, the research progress of taxanes nanostructures in recent years is reviewed in order to provide reference for the development and application of new pharmaceutical dosage forms.
