目的观察重型再生障碍性贫血(简称SAA)患者外周血CD4+CD25+调节性T细胞转录因子FOXP3的表达水平及白细胞介素10(IL-10)的分泌水平。方法选用6例初发SAA、6例免疫治疗后造血恢复的SAA患者及8例健康对照的外周血标本。免疫磁珠分选CD4+CD25+T细胞,分别用ELISA法和RT-PCR法检测CD4+CD25+T细胞培养后IL-10分泌水平和转录因子FOXP3mRNA的表达水平。结果初发SAA组IL-10分泌水平和转录因子FOXP3mRNA的表达水平均显著低于造血恢复组和正常对照组(P<0.01),后2组间无明显差异。结论IL-10和转录因子FOXP3与SAA的发病可能有关。
Objective To investigate the expression of CD4 + CD25 + regulatory T cell transcription factor FOXP3 in peripheral blood and the level of interleukin 10 (IL-10) secretion in patients with severe aplastic anemia (SAA). Methods Six cases of primary SAA, six cases of SAA with hematopoietic recovery after immunotherapy and eight healthy controls were selected. The CD4 + CD25 + T cells were sorted by immunomagnetic beads. The secretion of IL-10 and the expression of FOXP3 mRNA in CD4 + CD25 + T cells were detected by ELISA and RT-PCR respectively. Results The level of IL-10 secretion and the expression of FOXP3 mRNA in SAA group were significantly lower than those in hematopoietic recovery group and normal control group (P <0.01). There was no significant difference between the two groups. Conclusion IL-10 and the transcription factor FOXP3 may be involved in the pathogenesis of SAA.
