采用 5种分离培养基从油菜和芹菜的茎和叶中分离得到 2 15株内生菌 ,通过菌块对峙法对其拮抗植物病原真菌的性能进行初步的测定 ,并对发酵液的拮抗活性进行了进一步的筛选。结果表明 ,对蔬菜立枯丝核菌(Rhizoctoniasolani)、直喙镰孢菌 (Fusariumorthoceras)、苹果黑腐皮壳 (Valsamali)有拮抗作用的菌株有 18株 ,包括10株细菌和 8株放线菌。分离自油菜茎的放线菌CHSH19A有极强的抗立枯丝核菌活性 ,通过显微镜和电镜下的观察 ,初步判断为链霉菌 ;进而对其进行DNA提取 ,16SrDNA全长扩增和长度为 1382bp的序列测定及序列分析证明 ,CHSH19A为一株链霉菌Streptomycessp .
A total of 2 15 strains of endophytes were isolated from the stems and leaves of canola and celery using five kinds of isolation media. The antibacterial activities against phytopathogenic fungi were determined by the confluence method. The antagonistic activity of the fermentation broth For further screening. The results showed that there were 18 strains of antagonistic bacteria against Rhizoctonia solani, Fusariumorthoceras and Valsamali, including 10 strains of bacteria and 8 strains of actinomycetes . Actinomycete CHSH19A isolated from the stem of rapeseed has strong anti-Rhizoctonia solani activity, through the microscope and electron microscope observation, initially identified as Streptomyces; further its DNA extraction, 16SrDNA full-length amplification and length of 1382bp sequence analysis and sequence analysis proved that CHSH19A was a Streptomycessp.
