目的 探讨体外培养胎鼠下丘脑CRH神经元的形态、生长变化规律及CRH的表达变化。方法 取 17d胎鼠下丘脑分散培养下丘脑神经元 ,镜下连续观察神经元胞体直径、突起长度、数目等变化规律 ,应用免疫组织化学染色方法观察CRH神经元的生长变化规律。结果 培养下丘脑CRH神经元 3~ 6d生长速度最快 ;7~ 10d神经元细胞数量、胞体直径、突起长度和CRH染色强度均达高峰 ;12d以后部分神经元出现退形性改变。结论 培养下丘脑CRH神经元 7~ 10d迅速生长趋于成熟 ,是神经元机能测试的最佳时期
OBJECTIVE: To investigate the morphological changes of CRH neurons in fetal rat hypothalamus and the changes of CRH expression. Methods Hypothalamus neurons were disseminated in the hypothalamus of rats for 17 days. The changes of cell diameter, length and number of neurons were observed under microscope. The growth of CRH neurons were observed by immunohistochemical staining. Results The neurons in hypothalamus cultured for 3 ~ 6 days had the fastest growth rate. The number of neurons, the diameter of body cells, the length of protuberances and the intensity of CRH staining peaked at 7 ~ 10 days. After 12 days, some neurons showed degenerative changes. Conclusion The rapid growth of CRH neurons in hypothalamus 7 to 10 days tends to mature and is the best period for neuronal function test
