目的 克隆维甲酸诱导的前列腺癌细胞 DU- 145凋亡的未知相关基因 .方法 以全反式维甲酸 (all- trans retinoicacid)诱导前列腺癌细胞系 DU- 145细胞凋亡 ,在此基础上 ,采用基于 PCR技术的改良消减杂交方法克隆前列腺癌凋亡相关基因 .结果 成功克隆出一条与凋亡可能有密切关系的未知基因 pcar,已被 Gen Bank收录 ,登录号为 AF174394.结论 维甲酸诱导前列腺癌细胞 DU- 145凋亡的过程是一个多基因参与的复杂过程 ,pcar可能是参与这一过程的基因之一
OBJECTIVE: To clone the unknown related genes of retinoic acid-induced apoptosis of prostate cancer cell line DU-145.Methods Apoptosis of prostate cancer cell line DU-145 cells was induced by all-trans retinoic acid.On this basis, Cloning of apoptosis-related genes in prostate cancer by PCR-based improved subtractive hybridization.Results A novel gene pcar, which may be closely related to apoptosis, has been successfully cloned and has been deposited with Gen Bank under accession number AF174394.Conclusion Retinoic acid-induced prostate cancer The process of cell DU-145 apoptosis is a complex process involving multiple genes, and pcar may be one of the genes involved in this process
